To turn on or off Tamper Protection for Microsoft Defender Antivirus, just follow the next steps:

First, using the key combination Win + I, launch the Settings application, and in it go to Privacy and Security, in which you should click on Windows Security.
In the window that opens, click on Virus and threat protection, and then go to Manage settings.
At the bottom of the page that opens, you will find the Tamper protection switch, which will allow you to enable or disable protection from unauthorized access in Microsoft Defender.
